    Carum carvi, commonly known as caraway, is a medicinal and culinary plant recognized for its anti-inflammatory properties, primarily attributed to its essential oil components. However, the thermogenic potential of caraway-particularly the biological activity of its water-soluble extract-remains largely unexplored. In this study, we investigated the effects and underlying mechanisms of caraway on Ucp-1 mRNA expression in beige adipocytes and on inflammation-mediated suppression of thermogenesis, by treating C3H10T1/2 adipocytes with caraway water extract (CWE) or caraway hexane extract (CHE) during both the induction and maturation phases, followed by isoproterenol stimulation, and measurement of mRNA levels of Ucp-1 and differentiation-related genes. Additionally, RAW264.7 cells were treated with CWE prior to stimulation with lipopolysaccharides followed by evaluation of inflammatory marker expression. CWE increased Ucp-1 mRNA expression directly by enhancing adrenergic sensitivity and promoting beige adipocyte differentiation during the induction phase of differentiation. Further, CWE mediated an indirect effect on Ucp-1 expression by suppressing macrophage inflammation, thus restoring Ucp-1 expression otherwise inhibited under inflammatory conditions. These results suggest that caraway extracts-especially the water-soluble compounds-may serve as therapeutic candidates for obesity-related conditions by enhancing energy expenditure and mitigating chronic inflammation.

    Abstract

    We investigated the effects of piceatannol on the osteogenic differentiation of human periodontal ligament fibroblasts (HPLFs). Treatment with piceatannol increased alkaline phosphatase activity and osteogenic differentiation gene molecules (Osteocalcin and Runx2), but this was attenuated by mitogen-activated protein kinase (MAPK) inhibitors (PD098059 or SB203580). Our data suggest that piceatannol promotes osteogenic differentiation of HPLFs by activating MAPKs.

    Abstract

    Recently, it has been suggested that brown and beige adipocytes may ameliorate obesity because these adipocytes express uncoupling protein-1 (UCP-1), which generates heat by consuming lipid. However, obesity-induced inflammation suppresses the expression of UCP-1. To improve such conditions, food components with anti-inflammatory properties are attracting attention. In this study, we developed a modified system to evaluate only the indirect effects of anti-inflammatory food-derived compounds by optimizing the conventional experimental system using conditioned medium. We validated this new system using 6-shogaol and 6-gingerol, which have been reported to show the anti-inflammatory effects and to increase the basal expression of UCP-1 mRNA. In addition, we found that the acetone extract of Sarcodon aspratus, an edible mushroom, showed anti-inflammatory effects and rescued the inflammation-induced suppression of UCP-1 mRNA expression. These findings indicate that the system with conditioned medium is valuable for evaluation of food-derived compounds with anti-inflammatory effects on the inflammation-induced thermogenic adipocyte dysfunction.

    We previously established trophoblast stem cells from mouse androgenetic embryos (AGTS cells). In this study, to further characterize AGTS cells, we compared cell proliferation activity between trophoblast stem (TS) cells and AGTS cells under fibroblast growth factor 4 (FGF4) signaling. TS cells continued to proliferate and maintained mitotic cell division in the presence of FGF4. After FGF4 deprivation, the cell proliferation stopped, the rate of M-phase cells decreased, and trophoblast giant cells formed. In contrast, some of AGTS cells continued to proliferate, and the rate of M-phase cells did not decrease after FGF4 deprivation, although the other cells differentiated into giant cells. RO3306, an ATP competitor that selectively inhibits CDK1, inhibited the cell proliferation of both TS and AGTS cells. Under RO3306 treatment, cell death was induced in AGTS cells but not in TS cells. These results indicate that RO3306 caused TS cells to shift mitotic cell division to endoreduplication but that some of AGTS cells did not shift to endoreduplication and induced cell death. In conclusion, the paternal genome facilitated the proliferation of trophoblast cells without FGF4 signaling.

    これまでに我々は、幼若期(被験動物)における鉄欠乏状態が骨脆弱化の誘導、細胞老化亢進や細胞機能に多様性を付与するタンパク質のメチル化状態が低下することを報告している。加えて、鉄欠乏状態が活性型ビタミンD3やビタミンC律速酵素SMP30発現低下を惹起することも明らかにしている。興味深いことに、脳機能低下が認められるうつ病患者は、鉄をはじめとしたミネラル類、ビタミンD3やビタミンCといった栄養素が不足していることも明らかにされつつある。しかしながら、鉄摂取不足により脳機能の低下が増加するのか?または脳機能の低下を患った結果なのか?については明らかではない。そこで本年度は成長期における鉄欠乏食投与が脳機能に関わるタンパク質が変化するのかについてショットガン解析を用いて網羅的に解析を行った。
    被験動物として、4週齢のICR雄マウスを用い、正常食(CTL)と鉄欠乏群(ID)の計2群に分け、4週間の飼育観察を行った。飼育期間終了後、血中ヘモグロビン濃度の著しい低下、心臓および脾臓重量の有意な増加を確認し、鉄欠乏モデルであることを確認した。また、脳重量および脳内鉄濃度について解析した結果、2群間に有意な差を示さなかった。続いて、脳内タンパク質の変動をLC-MS/MSベースのプロテオミクス解析で行った結果、CTL群に比し、ID群で196因子の発現が減少(log2FC≦-0.5)を示した。特に有意に発現が減少した因子として、鉄がタンパク質活性に関連する因子や脳神経発達に関連する因子であった。加えて、トリプトファン代謝に関するタンパク質発現の多くがID群で減少していることも併せて明らかにした。
    以上より、鉄欠乏状態において脳内鉄濃度に変化は認められないものの、脳機能に影響を及ぼすタンパク質が有意に低下するプロファイリングを示し、鉄欠乏状態が脳機能に影響を及ぼす可能性を示唆した。

    本研究ではビタミンDが子宮内膜間質細胞の脱落膜化に与える影響を解明することを目的とした。研究初年度は以下の通り取り組んだ。
    ・in vitroにおける子宮内膜間質細胞の脱落膜化の再現
    ヒト子宮内膜間質細胞由来の不死化細胞株であるKC02-44D細胞は脱落膜化を再現できる細胞として報告されている。そこでKC02-44D細胞をエストロゲン存在下で4日間培養した後、プロゲステロン、ジブチリルcAMPを加え、8日間培養することで脱落膜化の誘導を試みた。プロゲステロン、ジブチリルcAMP添加時を脱落膜化誘導後0日目とし、0、4、8日目に脱落膜化の指標となるPRL遺伝子、IGFBP1遺伝子のmRNA発現をPCR法により解析した。結果として、どちらの遺伝子も誘導後8日目にmRNA発現が認められた。また、脱落膜化を促す3種の転写因子のmRNA発現も脱落膜化誘導後0、4、8日目に認められた。以上の結果から、KC02-44D細胞を用いることでin vitroにおいて子宮内膜間質細胞の脱落膜化が再現できると考えた。また、脱落膜化誘導過程における水酸化酵素CYP27B1並びにビタミD受容体の発現解析を行った。ビタミンDの活性化に関与するCYP27B1、並びにビタミンD受容体は子宮内膜において発現することが報告されている。本研究に用いるKC02-44D細胞において、これらの遺伝子が発現しているかPCR法により解析した。結果として、脱落膜化誘導後0、4、8日目の細胞においてCYP27B1、並びにビタミンD受容体のmRNA発現が認められた。
    以上の結果から、KC02-44D細胞は脱落膜化の過程におけるビタミンDの作用機序を明らかにする上で対象試料として適切であると考えた。
