    Exogenous oxidized cholesterol (OxChol) has deleterious effects on lipid metabolism. We therefore examined the effect of simultaneous consumption of wine polyphenol (WPP) and fish oil containing a high level of docosahexaenoic acid (DHA) in rats fed OxChol. The inhibition of hepatic cholesterol biosynthesis by exogenous OxChol was slightly alleviated by the consumption of both WPP and DHA. The activity of hepatic cholesterol 7α-hydroxylase tended to be higher in rats fed both WPP and DHA than in those fed the control diet without polyphenol containing safflower oil as dietary fat. On the other hand, dietary WPP and DHA lowered hepatic Δ46 desaturase activity compared with that in the control group. Moreover, the level of OxChol excreted in feces was significantly higher in rats fed both WPP and DHA than in those fed the control diet. Thus, the simultaneous consumption of both WPP and DHA seems to alleviate the modulation of lipid metabolism by dietary OxChol, possibly through a combination of both the inhibition of OxChol absorption from the small intestine by WPP and the modulation of lipid metabolism by DHA.

    Acidolysis and interesterification catalyzed by immobilized Lipase TOYO (<I>Chromobacterium viscosum</I>) in hexane have been studied to find a means of incorporating icosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) into triolein using free fatty acids and their ethyl esters as substrates. The fatty acids (EPA and DHA) were incorporated at about 40 %, while their esters only at about 10 %. The acidolysis of fish oil (sardine oil and yellowfin tuna eye socket oil) with EPA and DHA gave <I>n</I>-3-polyunsaturated fatty acid (PUFA) rich triglyceride (EPA+DHA incorporation higher than 60 %). The immobilized enzyme may thus be considered useful for producing <I>n</I>-3-PUFA rich oil.

    Enzymatic glyceride synthesis and acid interchange using icosapentaenoic acid (EPA), docosahexaenoic acid (DHA), γ-linolenic acid (GLA), and EPA and DHA concentrated saponified fatty acid mixture obtained from sardine oil were carried out through the use of four kinds of microbial lipases.<br> Lipase TOYO (<i>Chromobacterium viscosum</i>) was the most effective enzyme for glyceride synthesis as well as acid interchange of triglyceride (TG) rich in polyunsaturated fatty acids. There was a general tendency, except with Lipase TOYO, that DHA was difficult to be incorporated into a glycerol molecule compared to the other fatty acids.<br> Fatty acid composition of TG synthesized from a highly concentrated fatty acid mixture (EPA+DHA: 68.8%) by the use of Lipase TOYO and Lipase OF (<i>Candida cylindracea</i>), directly reflected that of the substrate fatty acid flixture (i.e. EPA+DHA was more than 65%). And the TG molecular species having combination of only EPA and DHA such as (EPA, EPA, EPA), (EPA, EPA, DHA), (EPA, DHA, DHA), and (DHA, DHA, DHA) were dominant accounting for the half of the obtained TG.

    The rates of incorporation into triolein of eight free fatty acids [palmitic acid (16 : 0), linoleic acid (18 : 2), α-linolenic acid (18 : 3), γ-linolenic acid (18 : 3), octadecatetraenoic acid (18 : 4), arachidonic acid (20 : 4), icosapentaenoic acid (20 : 5) and docosahexaenoic acid (22 : 6)] by acidolysis were determined. Lipase TOYO (<I>Chromobacterium viscosum</I>) and Lipozyme (<I>Mucor miehei</I>) both as immobilized lipase were used for the reactions.<BR>Lipase TOYO incorporated each free fatty acid into triolein at more than 22%. Lipozyme incorporated each free fatty acid at above 15%, and showed especially high activity toward icosapentaenoic acid.<BR>These two lipases were thus concluded applicable to the acidolysis of fat with unsaturated fatty acids.

    The hydrolysis and synthesis of icosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) rich oil by Lipase TOYO (<I>Chromobacterium viscosum</I>) were conducted.<BR>EPA and DHA were found to be released from the glycerol moiety at a constant rate, and were incorporated, by the reverse reaction, into the glycerol moiety using Lipase TOYO.<BR>The optimum water content and enzyme amount for synthesis of triglyceride were 0.93% and 200 units respectively. Under these conditions, using pure EPA as well as DHA as the substrate, 91. 3% of EPA and 94. 9% of DHA were incorporated into the glycerol moiety.

    Determination was made of the rates of icosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) incorporation into triolein and sardine oil triglyceride (TG).<BR>Three kinds of lipases (Lipase OF from <I>Candida cyrindracea</I>, Lipase A from <I>Aspergillus</I> sp. and Lipase TOYO from <I>Chromobacterium viscosum</I>) were first immobilized onto Celite 545. A preliminary study was then performed to determine optimum conditions for acidolysis by conducting this process between triolein and palmitic acid, using these immobilized lipases including Lipozyme<SUP>TH</SUP> (<I>Mucor miehei</I> lipase was immobilized on a macroporous anion exchange resin). Optimum water contents for Lipase OF, Lipase A, Lipozyme, and Lipase TOYO reaction systems were 20.38%, 10.24%, 6.3810.38%, and 5.18%, respectively, and the optimum immobilized lipase amount was about 250 mg for any of the four lipases.<BR>Under the above conditions, the acidolysis of triolein between pure EPA and then DHA was carried out. EPA incorporation rate was found highest in the system using Lipase OF (25.09%), whereas in the case of DHA, it was highest in the system using Lipase TOYO (23.02%).<BR>Finally, acidolysis of sardine oil TG with an EPA, DHA concentrated free fatty acid mixture (EPA+DHA : 68.83%) was carried out and the results examined.<BR>As a result of the low incorporation rates of EPA and DHA into the original sardine oil TG, the percentage of DHA in the reactant TG doubled in the reaction systems using Lipase TOYO and Lipozyme.

    Pharmacokinetics and metabolism of cholesterol oxidation products (COP) was examined in this study. Dietary COP was transferred to peripheral tissue by incorporation in lipoprotein after absorption from intestine. The transitivity to brain of dietary COP was also observed. Metabolism of 7-ketocholesterol was different between male and female in rats. A part of COP may be excreted after glucuronic acid conjugation. Thus, a part of the metabolism of unknown COP was elucidated in this study. Moreover, food composition which prevented hazardousness of dietary COP was examined. Prenylated flavonoid, xanthohumol, inhibited COP absorption from intestine by modulation of gene expression of NPC1L1 and interference of simple diffusion. Therefore, hydrophobic flavonoids may inhibit hazardous of dietary COP.

    Dietary oxidized cholesterol was incorporated into lipoprotein as well as cholesterol and transported to peripheral tissues. 7-Ketocholesterol related to various diseases may be difficult to be metabolized with age. Therefore, the modulation in metabolism of 7-ketocholesterol may be related to the onset of a disease with an increased risk of developing with age.

    Hop (Humulus lupulus L.) pomace contains procyanidin-rich polyphenols, large oligomeric compounds of catechin. Effect of high dose (1%) of dietary hop pomace polyphenols (HP) in OLETF rats, an animal model of type 2 diabetes, was studied. The rats fed HP tended to have a lower body weight and reduced white adipose tissue weight than the rats fed a control diet. Triglyceride levels in both plasma and liver tended to be lower in the HP fed group than in the control group. Thus, dietary HP may exert anti-obesity effect through the ameliorative function on hepatic fatty acid metabolism and inhibition of fat and glucose absorption from intestine.

    In this study, I examined the high dose effects of procyanidins from apple polyphenol(AP), hop polyphenol(HP), and tea polyphenol(TP) on obesity using normal Sprague-Dawley rats on a high fat(HF) diet. I measured adipose tissue mass and tissue lipid parameters in rats on polyphenol-free low fat(LF)(7%) or HF(27.8%) diets with or without 1% of each polyphenol for 9 weeks. Dietary AP, HP, and TP suppressed the increase in white adipose tissue. Dietary TP increased the level of fatty acids excreted into feces ; however, this was not observed in the AP-or HP-fed group. Each dietary polyphenol tended to reduce fatty acid synthesis and promote fatty acid β-oxidation as compared with a HF diet alone. Moreover, each dietary polyphenol tended to increase the level of peroxisome proliferator-activated receptor alpha(PPARα) in the liver and decreased the levels of PPARγ in brown and white adipose tissues. Therefore, I speculate that procyanidins from AP or HP may reduce the increase in white adipose tissue induced by an HF diet through a combination of the agonist-like action of PPARa and antagonist-like action of PPARγ. On the other hand, TP may exert an anti-obesity effect via the combined effect of PPARα and PPARγ described above as well as the promotion of fatty acid excretion into feces. The dose-dependent hypocholesterolemic and anti-atherogenic effects of dietary AP from unripe apple, which contains approximately 85% procyanidins were also examined in male Sprague-Dawley rats(4 weeks of age) given a purified diet containing 0.5% cholesterol. Dietary AP at 0.5 and 1.0% levels significantly decreased the liver cholesterol level compared with that in the Control(AP-free diet-fed) group. Dietary AP also significantly lowered the serum cholesterol level compared with that in the Control group. However, the HDL cholesterol level was significantly higher in the 1.0% AP-fed group than in the Control group. Accordingly, the ratio of HDL-cholesterol/total cholesterol was significantly higher in the 0.5% AP-fed group and 1.0% AP-fed group than in the Control group. Moreover, the atherogenic indices in the 0.5% and 1.0% AP-fed groups were significantly lower than those in the Control group. The activity of hepatic cholesterol 7α-hydroxylase tended to be increased by dietary AP in a dose-dependent manner. In accord with this observation, dietary AP increased the excretion of acidic steroids into feces. Dietary AP also significantly promoted the fecal excretion of neutral steroids in a dose-dependent manner. These observations suggest that dietary AP at a 0.5 or 1.0% level exerts hypocholesterolemic and anti-atherogenic effects through the promotion of cholesterol catabolism and inhibition of intestinal absorption of cholesterol.

    コレステロール酸化物を含む純化飼料を8週齢のSD系雄ラットに与え、10日間飼育した結果、肝臓のHMG-CoA reductase活性ならびにcholesterol 7α-hydroxylaseのmRNA発現レベルは低下し、とくに、後者の発現は大きく低下し、それを反映して糞中に排泄される胆汁酸レベルも大きく低下した。一方、脂肪酸代謝に関しては、肝臓のdelta5 desaturaseおよびdelta5 desaturaseのmRNA発現レベルはコレステロール摂取群と無コレステロール食摂取群よりも有意に高くなった。これを反映し、両酵素活性は高くなり、脂肪酸組成変動にもその影響が観察された。また、核内受容体レベルの変化も観察され、コレステロール酸化物摂取による成長阻害あるいは中性脂肪レベルの大きな低下の原因を突き止めることができた。
    上記のような作用に対して、果実由来プロシアニジン、大豆イソフラボン、水溶性食物繊維、茶カテキン、植物性蛋白質の摂取によって、部分的に改善されること、あるいは、数種の食品成分のコンビネーションによって大きく脂質代謝攪乱が改善されることを見いだした。
    以上のように、加工食品全盛に時代にあって、多食されている可能性が高いコレステロール酸化物による脂質代謝攪乱作用の機序とその防止策としての食事コンビネーションを提示する基礎データを得ることができた。

    Biological activities of apple polyphenol oligomer derived from unripe apple in this study. Peroxidation of low density lipoprotein was strongly inhibited by apple polyphenol oligomer in in vitro assay and its potency was equal to that of tea catechin. Dietary apple polyphenol oligomer lowered the levels of liver phospholipids hydroperoxides in rats in the dose-dependent manner. Dietary apple polyphenol oligomer also lowered the activities of hepatic HMG-CoA reductase and Δ6 desaturase in rats. On the other hand, the activity of hepatic cholesterol 7α-hydroxylase was increased by dietary apple polyphenol oligomer. Reflecting these modulations of enzyme activities, liver cholesterol level and Δ6 desaturation indices of tissue lipids were lowered and fecal excretion level of acidic steroids was increased in rats fed apple polyphenol oligomer. The ratio of HDL-cholesterol/total cholesterol was increased by intake of apple polyphenol oligomer The dimer and trimer of apple polyphenol oligomer were identified in serum in rats fed apple polyphenol oligomer. Moreover, many unidentified compounds derived from apple polyphenol oligomer were found in serum in rats fed apple polyphenol, although these compounds were not identified in serum in rats fed polyphenol-free diet. Therefore, absorbed and decomposed compounds derived from apple polyphenol oligomer may exhibit various biological functions such as antioxidative action and regulation of lipid metabolism.

    研究最終年度は、コレステロール過酸化物を作成かつ精製し、マクロファージあるいは動脈内皮細胞のエイコサノイド産生に与える作用を検討した。まず、マクロファージを細胞毒性がLD50以下でコレステロール過酸化物を添加した培養液で培養した結果、同じレベルのコレステロールに曝露した場合と比べると、アラキドン酸放出ならびにプロスタグランジンE2産生と放出量が高まることが明らかとなった。また、この原因として、COX-1の活性が高まること、その原因は酵素レベルが高まることも確認できた。すなわち、コレステロール過酸化物が生体内に存在すると、一方、動脈内皮細胞を同様にコレステロール過酸化物に曝露した結果、マクロファージと同様にコレステロール曝露では認められなかった。アラキドン酸放出の上昇とプロスタサイクリン産生濃度がまった。その原因には、細胞のホスホリパーゼA2とCOX-1の活性の上昇が主な原因であることを突き止めた。このように、生体内の過酸化コレステロールレベルが高まると、種々の炎症やエイコサノイドレベルのインバランスによる代謝障害の引き金になる可能性が考えられた。過酸化ステロールの有害作用をin vivoで確認するために、実験動物(ラット)にCCl4を投与し、組織の過酸化コレステロール濃度とエイコサノイドレベルを調べた。その結果、CCl4投与により、対照である無CCl4投与群と比較して、著しく組織過酸化コレステロール濃度は上昇した。さらに、動脈プロスタサイクリン産生濃度と腎臓のプロスタグランジンE2濃度も上昇した。このように、間接的ではあるが、in vitroでのコレステロール過酸化物による代謝撹乱が確認された。これらの結果を総括すると、食事由来を含めて、生体内に高濃度の過酸化コレステロールが存在すると、脂肪酸ならびにエイコサノイド代謝を撹乱し、動脈硬化を含む種々の疾病発症に至る可能性が予想された。この原因は、代謝を担う種々の酵素活性に変動を与えることが主要な原因であることもわかった。

    加工食品に無視できないレベルで含まれている酸化コレステロール(OxCHOL)が小腸から吸収されると、肝臓のCHOL生合成と異化を低下させ、一方ではリノール酸不飽和化を亢進することを見出した。このことから、食餌性OxCHOLは代謝諸機能を変動させて、種々の疾病の引き金になる可能性が懸念された。そこで、このようなOxCHOLの生理機能を抑制する食品成分を検索することに焦点を当てて、りんごポリフェノールによる制御機能を検討した。まず、りんごポリフェノールを0.5あるいは2.5%含む(対照は含まない)AIN76純化食にOxCHOLを0.3%加えた飼料をSD系雄ラット(各群7匹)に3週間与えた。飼育終了後、腹部大動脈より採血して屠殺し、肝臓ミクロソームを調製して、リノール酸不飽和化律速酵素のΔ6 desaturase活性を測定した。酵素活性は対照群に比べて、りんごポリフェノールを摂取した群では有意に低下した。また、この活性低下を反映して、血清ならびに肝臓脂質のリノール酸代謝産物/リノール酸は対照に比べてりんごポリフェノール摂取群で有意に低かった。この結果から、りんごポリフェノール食餌性OxCHOLが誘発するリノール酸不飽和化亢進作用を抑制するとともにエイコサノイド産生増大を制御してアレルギーや炎症反応を引き起こしにくくする可能性が考えられた。また、血清と肝臓の脂質成分ならびに過酸化脂質濃度を調べたところ、OxCHOL摂取で生じる変動はりんごポリフェノールの摂取で抑制された。りんごポリフェノールによる代謝調節機能の一因として食餌性OxCHOLの吸収制御が考えられたので、ミセルへのOxCHOL可溶化に対するポリフェノールの影響をin vitroで調べた。その結果、ポリフェノールは濃度依存的にOxCHOL可溶化を阻害した。このことから、OxCHOLが誘発する代謝変動に対するりんごポリフェノールの調節機能の一端はその吸収制御である可能性が推察された。
