    The C5-C11 moiety of phoslactomycin I-i was synthesized via chelation-controlled addition of CH2═CHMgBr to the C8 ketone, ozonolysis, and the HWE reaction. The TMS acetylene and C1-C4 were attached to C11 and C5, respectively. A cyclohexyl moiety possessing an iodovinyl group was synthesized from quinic acid. The coupling reaction of the intermediates followed by Zn reduction yielded the cis,cis-diene. Finally, the amino and phosphate groups were attached.

    Abstract

    We studied the suppression of radioactive Cs elution from municipal solid waste incineration (MSWI) fly ash using 10-[(tert-butyldiphenylsilyl)oxy]-decanoic acid. The synthesized decanoic acid derivative coated on MSWI fly ash containing less than 8000 Bq kg<sup>−1</sup> of radioactive Cs, and the amount of radioactive Cs eluted into pure water was measured by the Japanese Leaching Test No. 13. The elution rate of <sup>137</sup>Cs from raw fly ash was normalized to 100%, and the relative elution rate of <sup>137</sup>Cs using 10-[(tert-butyldiphenylsilyl)oxy]-decanoic acid was 25.3%. The synthesized carboxylic acid showed a high elution suppression effect on <sup>137</sup>Cs in MSWI fly ash. SEM revealed that the water repellent layer was maintained on the MSWI fly ash treated with carboxylic acid synthesized around JLT-13. In addition, the contact angle measurement showed that the surface of the MSWI fly ash treated with the synthesized carboxylic acid for water repellency was hydrophobic. These results indicate that the surface of the MSWI fly ash was changed by applying a water repellent treatment with the synthesized carboxylic acid.

    Abstract

    We synthesized optically active (–)-cinmethylin with a high stereoselectivity. Sharpless asymmetric dihydroxylation of g-terpinene afforded an optically active diol with 98% ee. This diol was converted into (–)-cinmethylin in seven reaction steps. Dehydroxylation by the Barton–McCombie method was applicable, depending on the type of substituent, with the reaction affording the desired product when the substrate bore an OTBS group, but not when the substrate contained a benzyl ether. Decarbonylation gave the desired product regardless of the type of substituent. Furthermore, the stereoselectivity of the epoxidation reaction of an olefin with m-CPBA depended on the substituent, and up to 31% of the desired epoxide was obtained. Cyclization of the epoxide with PPTS proceeded with a good yield.

    Abstract

    A total synthesis of resolvin T4 was achieved by connecting three intermediates by Wittig reactions. The enal in the C1–C10 part was constructed through reduction of a propargylic alcohol with Red-Al followed by oxidation. The enal moiety in the C11–C16 part was synthesized by a ring-opening reaction of a silyl epoxide followed by a Peterson elimination. The chiral centers at C7 and C13 were constructed by ruthenium-catalyzed asymmetric transfer hydrogenation.

    Abstract

    Maresins are among the most potent antiinflammatory lipid metabolites. We report stereoselective syntheses of maresin 2 and maresin 2n-3 DPA. The anti-diol was constructed through epoxide ring opening of an optically active β,γ-epoxy aldehyde, synthesized in situ by Swern oxidation of the corresponding alcohol. Finally, the target compounds were synthesized through a Sonogashira coupling of a C9–C22 iodide and methyl (Z)-oct-4-en-7-ynoate or methyl oct-7-ynoate, respectively.

    The C16–C22 fragment with the acetylene terminus was constructed through the asymmetric dihydroxylation of the corresponding olefin, while the 15-iodo-olefin corresponding to the C11–C15 part was prepared via the asymmetric transfer hydrogenation of the corresponding acetylene ketone followed by hydrozirconation/iodination. Both pieces were joined by a Sonogashira coupling, and the product was further converted into the title compound via a Wittig reaction with the remaining C1–C10 segment and Boland reduction using Zn with TMSCl.

    ラクトマイシン類はStreptomyces sp. ACT232から単離された新規ポリケチド天然物である。ラクトマイシン類は強力なカテプシンB阻害活性をもち,新規な作用機序による抗がん剤のリード化合物として期待される。本研究では,ラクトマイシン類について,構造活性相関研究を志向した合成法を確立し,ラクトマイシン類やその誘導体・類縁体の合成を行う。
    2021年度はC8およびC9位の不斉中心を立体選択的に構築した後,C11位の不斉中心の構築について検討した。すなわち,1,4-ブタンジオールを出発物質として,10工程で合成したアルデヒドトリメチルシリルアセチレンを付加してC11位に相当する不斉中心を構築した。C9位の立体化学により立体選択的に進むと考えたが,混合物となった。そこで,得られたアセチレンアルコールをPCCにより酸化した後,ルテニウム触媒による不斉水素移動反応によりC11位の不斉中心を立体選択的に構築した。
    また,上述の方法は工程数が多く,不斉中心を立体選択的に構築することが難しい。そこで,C8位からC11位の3つの不斉中心をより簡便に,かつ立体選択的に構築するため,新しい合成法も並行して検討した。具体的には,C11位の不斉中心はグリシドールの立体化学を活用することとし,3-ブチン-1-オール誘導体をグリシドール誘導体に付加してC8からC13位炭素骨格を構築した。その後,数工程で3置換オレフィンを構築した。

    紡糸セルから射出させる方法でナノ繊維を得るアプローチについて、十分なゲル化速度が得られず、合成条件等の再検討が必要となった。反復凍結融解法を適用してコロイドナノ粒子を高分散した多孔性ゲル吸着剤を合成し、種々の条件における水分吸脱着データをバッチ系の水分吸着装置を用いて取得してきた。この手法で作成したゲル吸着材は非常に早い水分吸着特性を示し、予備研究で取得したナノ繊維の吸湿速度と同等の性能をしめした。一方で、この多孔質ゲルは脱着速度が早くなく、ゲル粒子の大きさに依存する拡散移動が律速していると考えられた。合成したナノコロイド分散多孔質ゲルについては、ゲル構造の評価方法及び分析について、外部の企業の協力を得て実施した。SEM分析により、コロイド粒子が高分散で固定したゲル細孔表面が確認できた。用いるコロイド粒子は分散粒子と比べ、ある程度連結したコロイドにおいて高分散固定できることをSEM分析により確認した。吸着データについては、作成した非定常拡散吸着モデルのシミュレーション結果との整合を検証した。これらの実験的な検討とは別に、ゲルを用いた吸着材について、新たに、直近10年間の論文をデータベースで検索してまとめた。調湿性能の評価を精度良く迅速に行うことを目的として新たに吸脱着特性測定装置を構築した。今年度はコロナ禍の影響もあり、研究推進に多くの支障をきたした。このため、研究期間の延長を申請した。

    We have investigated isomerization properties of [14]parapyridinophane derivatives and demonstrated that crystallization-induced and adsorption-induced asymmetric transformations are efficient protocols to prefer the opposite sense of stereochemistry in planar-chiral [14]parapyridinophane derivatives, respectively. We have also found that azaspirene analog having its characteristic spiro skeleton readily undergoes racemization in neutral aqueous media and that efficient resolution was achieved by using an appropriate chiral auxiliary to resolve the analog by the following acidic hydrolysis of each diastereoisomer to afford highly enantio-enriched azaspirene analog in excellent yields without a loss of their enantiomeric excesses. The sequential transformation to enantiomerically pure analog and its recycling process to its racemic body achieved an efficient synthetic route to an optically active analog of azaspirene.
