    Optimizing fruit quality is a key objective in controlled-environment agriculture for high-value crops, including strawberries. Fruit quality is largely determined by sweetness and aroma, both of which are strongly influenced by environmental conditions during fruit development, particularly water availability and light quality. However, the interactive effects of water deficit and far-red (FR) radiation on fruit yield and aroma composition remain insufficiently understood. We investigated the individual and combined effects of water deficit and FR radiation on ‘Tochiotome’ strawberry fruit development in a controlled growth chamber over a 3-year period. Treatments included water deficit alone, FR radiation alone, and their combined application. Fruit fresh weight, soluble sugar content, and volatile aroma compounds were analyzed using gas chromatography–mass spectrometry. Reduced irrigation levels decreased soil water potential significantly, confirming the induction of water deficit in strawberry plants. Water deficit resulted in a pronounced, threshold-dependent reduction in fruit fresh weight, accompanied by an increase in soluble sugar content. FR radiation alone had a comparatively minor effect on yield, but contributed to increased sugar accumulation, particularly under combined conditions. Notably, the combination of FR radiation and moderate water deficit reduced fruit weight significantly. Aroma analysis revealed that strawberry volatile profiles were dominated by esters and terpenoids, with (E)-2-hexenyl acetate, hexyl acetate, (E)-nerolidol, and linalool consistently identified as major components. Water deficit primarily induced quantitative changes and stress-related shifts in volatile composition, whereas FR radiation influenced metabolic pathway allocation, promoting terpenoid synthesis. Under combined treatments, water availability emerged as the dominant factor shaping aroma profiles, whereas FR radiation acted as a modulatory factor. Overall, water deficit was the primary determinant of fruit quality, whereas FR radiation played a secondary, modulatory role. These findings provide a basis for optimizing irrigation and light conditions to improve yield, sweetness, and aroma in controlled-environment strawberry production.

    Purple paprika ( Capsicum annuum ) is a vegetable with potential economic value; however, enhancing and maintaining the purple fruit color is challenging. We investigated the effects of night blue-light supplementation on fruit growth, anthocyanin content, and gene expression in purple paprika. During two duplicated experiments conducted in spring and autumn, purple paprika plants were subjected to blue-light supplementation at night. Ten days after fruits were pollinated, night blue light with photosynthetic photon flux density (PPFD) of 80 to 100 µmol·s <sup>−1</sup> ·m <sup>−2</sup> was supplied from 1800 HR to 0500 HR (11 hours). Fruit samples were harvested 15, 20, and 40 days after pollination from control (no-light treatment) and blue light–treated fruits. The fruit size, fresh weight, anthocyanin content, and expression levels of anthocyanin biosynthesis-related genes ( HY5 , bHLH , WDR , MYB , PAL , CHS , F3H , DFR , ANS , and UFGT ) were determined. Blue-light supplementation increased the anthocyanin-related gene expressions in the fruit peel, enhancing anthocyanin synthesis and accumulation. However, there were no significant differences between the growth of control and blue light–treated fruits. These findings highlight the potential of night blue-light supplementation to enhance anthocyanin content in purple paprika fruits without affecting overall fruit growth.

    We investigated sugar (solute) accumulation in watermelon [ Citrullus lanatus (Thunb.) Matsum. et Nakai] fruits at the immature stage. Watermelon plants were grown hydroponically in a nutrient solution with an electric conductivity (EC) of 1.2 S⋅m <sup>−1</sup> (EC 1.2 regime); then, fruits were harvested 21 days after anthesis. The flesh of each fruit was divided into seven different parts to measure the sugar concentration and water status. The results indicated that the sugar concentration was higher in the center of the fruit flesh than in the other parts, such as around the pericarp. Moreover, the lowest osmotic potential was observed in the center of the fruit flesh, indicating solute accumulation. Concurrently, when the transport of photosynthates in the fruit was investigated using the <sup>13</sup> CO <sub>2</sub> isotope, the active solute accumulation in the center of the fruit flesh was observed, supporting the observed sugar accumulation in this part. Consequently, this active solute accumulation and distribution occurred in the center of the watermelon fruit, as demonstrated by the data of osmotic pressure and sugar concentration and supported by the observed active photosynthate accumulation. Additionally, we investigated these measurements by increasing the nutrient solution concentration 14 days after anthesis. As a result, fruit growth was slightly inhibited using the EC 3.0 regime, and <sup>13</sup> C translocation was also inhibited in the fruit, especially in its center. Even though the sugar concentration and osmotic pressure of the fruit flesh were not clearly affected by high nutrient solution concentrations, the cell turgor of the central flesh of the fruit grown using the EC 2.0 and 3.0 regimes was lower than that of the fruit grown using the EC 1.2 regime. Treatments with higher nutrient concentrations might have negative effects on immature watermelon fruits.

    During development, the fruit of some paprika ( Capsicum annuum L.) cultivars shows a change in color from green to dark purple (e.g., ‘Mavras’) or lilac (e.g., ‘Tequila’). However, this purple coloration is rare among paprika cultivars and disappears in ripened fruit, which are red. Therefore, we investigated the mechanism causing this color change in the cultivars Mavras and Tequila to better understand how purple ripened fruit could be generated. High-performance liquid chromatography (HPLC) analyses of the anthocyanin contents of the fruit indicated that anthocyanin was undetectable in green fruit, accumulated in dark purple or lilac ones, and then decreased again in red ones in both cultivars. Furthermore, expressions of most of the analyzed anthocyanin biosynthesis–related genes and genes for their transcription factors increased in dark purple or lilac fruit and decreased in red ones, i.e., it was synchronized with the changes in anthocyanin contents. Furthermore, anthocyanin degradation activity as a result of peroxidases was detected at all stages but increased when the lilac or dark purple color started to fade. Thus, the development of purple coloration is caused by increased anthocyanin biosynthesis, whereas the fading of this coloration is a result of both a decrease in anthocyanin biosynthesis and an increase in anthocyanin degradation. At the ripening stage, the green pigment (chlorophyll) contents decreased, whereas the red pigment (carotenoid, particularly capsanthin) contents increased. However, these timings did not completely coincide with the timing of anthocyanin degradation, suggesting that the content of each pigment is individually regulated, and so purple, green, and red coloration could be freely expressed in mature paprika fruit.

    We investigated physiological differences in watermelon [ Citrullus lanatus (Thunb.) Matsum. et Nakai] fruits among seeded diploid and seedless triploid fruits, N-(2-chloro-4-pyridyl)-N′-phenylurea (CPPU)–treated seedless fruits, and soft-X–irradiated pollen-pollinated seedless fruits to investigate the effect of the presence or absence of seeds on water relations and sugar content. We picked fruits at 20 and 40 days after anthesis and sampled flesh at the center, around the seeds, and near the pericarp to measure water status and sugar content. There were no significant differences between seeded and seedless cultivars in sugar contents or in water and osmotic potentials of the flesh, although the latter two were decreased at 40 days. CPPU and soft-X–irradiated pollen eliminated mature seeds, but there were again no significant differences in sugar contents or water status between seeded and seedless fruits. Thus, the presence or absence of seeds did not influence the sugar content or osmotic pressure in watermelon fruit, so sugar accumulation was not related to seeds.

    We investigated the effect of sugar and salt application to hydroponic culture solution for coloring red-leaf lettuce (<I>Lactuca sativa</I> L. 'Prunai' and 'Rouxai') under controlled culture conditions. In plant factories condition, red-leaf lettuce varieties frequently do not turn red. In order to solve this, we tested various stress conditions. Stresses were induced by addition of polyethylene glycol, sugar and salt to hydroponic culture solution, and using concentrated nutrient solution. The result showed that the combination of adding sugar and salt (5 and 0.02 % respectively in the nutrient solution at EC 0.6dSm<SUP>-1</SUP>) accerelated coloring of red-leaf lettuce varieties. This technique might be efficient for growing red-leaf lettuce cultivation in plant factories.

    We demonstrated the effect of cooling of the medium on the fruit set of strawberries ( Fragaria × ananassa Duch.) grown on high benches for forcing culture. The cooling by water evaporation promoted by a fan enabled to cool the medium by an average of several degrees compared with no cooling. When runner plants were transplanted in late summer, cooling accelerated flower bud emergence almost 10 days on the primary axillary branch compared with plants grown in uncooled medium. Also, with cooling, fruit was harvested from the inflorescence of the primary axillary branch almost 10 days earlier. We expect that this technique will allow early transplanting around the end of summer and will shorten the time between fruit set on the terminal inflorescence and that on the inflorescence of the primary axillary branch.

    This work was undertaken to investigate response of callus and the water status of Sturt's desert pea (<I>Swainsona formosa</I>) in vitro under various environmental stress conditions. Water potential of culture media ranged from -0.27MPa to -1.05MPa so that salt and osmotic stress and different concentration of sucrose could be applied to Sturt's desert pea calli grown in the tissue culture media. The percent increase in calli was highest in standard MS medium with 30g L<SUP>-1</SUP> sucrose but was not so when the water potential of medium was adjusted to the same water potential using standard MS plus mannitol. The water potential of callus tissue was almost the same as that of culture media at any range of MS and sucrose concentrations. Turgor of callus tissue did not alter significantly under any stress conditions. It is concluded that the optimum concentration of salt, sucrose and water status of medium could be estimated by this method and that standard MS concentration and its water status are suitable for the formation of Sturt's desert pea calli.

    Effects of light break treatment every second or third night on the growth of certain leaf vegetables were evaluated. In experiment 1, spinach (<i>Spinacia oleracea</i>) was irradiated with 0.4~1.2 μmol·m<sup>−</sup><sup>2</sup>·s<sup>−</sup><sup>1</sup> (PPF) of light from an incandescent lamp for 51 minutes a mid-night every night (every night treatment), or for two hours at mid-night on Monday, Wednesday and Friday (interval treatment). In experiments 2, spinach and tsukena (<i>Brassica campestris</i>) received interval treatment with 1.0-1.5 μmol·m<sup>−</sup><sup>2</sup>·s<sup>−</sup><sup>1</sup> of light of incandescent lamp for two hours at mid-night. In experiment 3, spinach, lettuce (<i>Lactuca sativa</i>) and tsukena received interval treatment with 0.5-1.8 μmol· m<sup>−</sup><sup>2</sup>·s<sup>−</sup><sup>1</sup> of light from light emitting diodes (LEDs) for two hours at mid-night.<br> In experiment 1, under interval treatment, there was no bolting of spinach on the 26<sup>th</sup> day of treatment. However, ratio of bolting plants was 22 % under every night treatment. In experiments 2 and 3, interval treatment with incandescent lamp or LED promoted spinach growth without bolting. In spinach, plant height, number of leaves and top dry weight were larger under interval light break treatment than under no light break. However, the growth of lettuce and tsukena were not promoted with light break treatment.<br>

    Cucumber (Cucumis sativus L.) seedlings were subjected to altered red/far-red photon flux ratio to investigate how it affected their growth and water status. Stem elongation of seedlings was hastened when exposed to the 0.73 of red/far-red photon flux ratio but it was inhibited when the ratio was 1.88. The growth-induced water potential between the regions of elongation and maturation of the seedlings grown under the ratio of 0.73 was 0.09 MPa. However, the growth-induced water potential was 0.03 MPa when seedlings were grown under ratio 1.88.

    We investigated water relations of fruit cracking in single-truss tomato (<I>Lycopersicon esculentum</I> Mill.) plants grown hydroponically in a greenhouse. At 24 d after anthesis, the water potential of the culture solution was changed from -0.06 MPa (control plants) to -0.36 MPa (stressed plants) . The water status of the culture solution and plant tissues was measured with an isopiestic thermocouple psychrometer. Fruit cracking decreased from>80% of fruit in the control to <30% in the stressed plants. The water potential gradient between fruit flesh and water source was greater in the control plants than in the stressed plants. Between predawn and morning, turgor increased in the fruit flesh in the control plants but not in the stressed ones. The hardness of the fruit skin was not different between stressed and control plants. These results indicate that the water potential gradient and increased turgor in the tissues might trigger fruit cracking in single-truss tomato plants.

    A new tissue culture medium was developed by mixing colloidal buffer agents together with tissue culture medium solution. Powders of zeolite, bentonite and charcoal were added to the medium solution as colloidal buffer agents. By adding colloidal buffer agents, the buffering capacity of pH in the medium increased significantly against both acidification and alkalization. When embryos taken out from <I>Phaseolus vulgaris</I> seeds were grown in media containing colloidal buffer agents, the length of plantlets formed from the embryos became larger and the dry weight of the plantlets became heavier than plantlets grown in media containing no buffering agents. This suggests that colloidal buffer agents stabilize the pH of the culture medium and sustain cell elongation and growth of plantlets for growing period during tissue culture. Furthermore, the addition of colloidal buffers in culture media had an additional benefit of preventing the degradation of organic substances in the culture media while the media were heated for the purpose of sterilization.

    This work was undertaken to investigate response of callus and the water status of Sturt's desert pea (<I>Swainsona formosa</I>) in vitro under various environmental stress conditions. Water potential of culture media ranged from -0.27MPa to -1.05MPa so that salt and osmotic stress and different concentration of sucrose could be applied to Sturt's desert pea calli grown in the tissue culture media. The percent increase in calli was highest in standard MS medium with 30g L<SUP>-1</SUP> sucrose but was not so when the water potential of medium was adjusted to the same water potential using standard MS plus mannitol. The water potential of callus tissue was almost the same as that of culture media at any range of MS and sucrose concentrations. Turgor of callus tissue did not alter significantly under any stress conditions. It is concluded that the optimum concentration of salt, sucrose and water status of medium could be estimated by this method and that standard MS concentration and its water status are suitable for the formation of Sturt's desert pea calli.

    We investigated water relations of fruit cracking in single-truss tomato (<I>Lycopersicon esculentum</I> Mill.) plants grown hydroponically in a greenhouse. At 24 d after anthesis, the water potential of the culture solution was changed from -0.06 MPa (control plants) to -0.36 MPa (stressed plants) . The water status of the culture solution and plant tissues was measured with an isopiestic thermocouple psychrometer. Fruit cracking decreased from>80% of fruit in the control to <30% in the stressed plants. The water potential gradient between fruit flesh and water source was greater in the control plants than in the stressed plants. Between predawn and morning, turgor increased in the fruit flesh in the control plants but not in the stressed ones. The hardness of the fruit skin was not different between stressed and control plants. These results indicate that the water potential gradient and increased turgor in the tissues might trigger fruit cracking in single-truss tomato plants.

    Cucumber (Cucumis sativus L.) seedlings were subjected to altered red/far-red photon flux ratio to investigate how it affected their growth and water status. Stem elongation of seedlings was hastened when exposed to the 0.73 of red/far-red photon flux ratio but it was inhibited when the ratio was 1.88. The growth-induced water potential between the regions of elongation and maturation of the seedlings grown under the ratio of 0.73 was 0.09 MPa. However, the growth-induced water potential was 0.03 MPa when seedlings were grown under ratio 1.88.

    A new tissue culture medium was developed by mixing colloidal buffer agents together with tissue culture medium solution. Powders of zeolite, bentonite and charcoal were added to the medium solution as colloidal buffer agents. By adding colloidal buffer agents, the buffering capacity of pH in the medium increased significantly against both acidification and alkalization. When embryos taken out from <I>Phaseolus vulgaris</I> seeds were grown in media containing colloidal buffer agents, the length of plantlets formed from the embryos became larger and the dry weight of the plantlets became heavier than plantlets grown in media containing no buffering agents. This suggests that colloidal buffer agents stabilize the pH of the culture medium and sustain cell elongation and growth of plantlets for growing period during tissue culture. Furthermore, the addition of colloidal buffers in culture media had an additional benefit of preventing the degradation of organic substances in the culture media while the media were heated for the purpose of sterilization.

    This study investigated the changes in endogenous auxin concentration during embryogenesis of citrus (Citrus sinensis Osb.) in tissue culture. Embryogenic callus (EC) was induced from ovules of 'Trovita orange' and 'Oomishima navel'. In both varieties, the EC having greater capacity for embryogenesis had a correspondingly higher endogenous indole-3-acetic acid (IAA) concentration. When the EC in 'Washington navel' and 'Oomishima navel' were cultured on hormone-free media which can induce embryogenesis, the IAA concentration increased for 2 days, then rapidly decreased and stabilized 14 days later. These results suggest that the decrease in IAA concentration is closely related to citrus embryogenesis.
