    Studies using functional magnetic resonance imaging assume that hemodynamic responses have roughly linear relationships with underlying neural activity. However, to accurately investigate the neurovascular transfer function and compare its variability across brain regions, it is necessary to obtain full-field imaging of both electrophysiological and hemodynamic responses under various stimulus conditions with superior spatiotemporal resolution. Optical imaging combined with voltage-sensitive dye (VSD) and intrinsic signals (IS) is a powerful tool to address this issue. We performed VSD and IS imaging in the primary (S1) and secondary (S2) somatosensory cortices of rats to obtain optical maps of whisker-evoked responses. There were characteristic differences in sensory responses between the S1 and S2 cortices: VSD imaging revealed more localized excitatory and stronger inhibitory neural activity in S1 than in S2. IS imaging revealed stronger metabolic responses in S1 than in S2. We calculated the degree of response to compare the sensory responses between cortical regions and found that the ratio of the degree of response of S2 to S1 was similar, irrespective of whether the ratio was determined by VSD or IS imaging. These results suggest that neurovascular coupling does not vary between the S1 and S2 cortices.

    新奇な物事に惹きつけられる度合いは,人によって,あるいは,年齢によっても異なるように見える.本研究では,こうした差異の基になる脳情報処理機構を調査するにあたり,動物(マウス)が新奇物体を好んで接近する行動に要する処理を,①新奇性への接近行動を開始させる処理,②新奇性の「評価と記憶」の処理,③好奇心のような感性が惹起する「報酬期待」の処理,の三種に分類する.その上で,各々のプロセスを担う神経基盤を調査する計画とした.
    (1)新奇性探索行動解析では,実験箱への順化の過程を,マウスの行動量により可視化し,環境順化に要する時間が,想定していた3日(10min/day)よりも長く必要である可能性が示唆された.また,DeepLabCutを活用してマウス(C57BL/6J)の鼻先が物体へ向いている状態を正確にトラックするツールを開発した.これを用いて,正方形 (40×40×40 cm)の実験箱で,10分間の二物体探索行動を解析した所,新奇物体へ積極的に接近する行動が,試験開始から約2分後に急激に消失する事が判明した.
    (2)探索行動中の脳活動計測のための,ファイバーフォトメトリー計測実験系を新たに構築し,ドーパミン(GRAB_DA)およびカルシウム(GCaMP)のシグナル計測に着手した.自由行動化のマウスで実際に計測したところ,ドーパミンやカルシウムの応答と思われるシグナルが検出できたものの,期待していた程の強さではなかった.組織学的な検証の結果,目的の部位にタンパク質が発現していたため,光ファイバーの埋込み位置に改善の余地があると結論した.
    (3)嗅周囲野・嗅内野における膜電位イメージング解析では,4-AP及びGanbazine 灌流下における刺激誘導性オシレーションのウェーブレット解析を行い,35野の特性が嗅内野の特性に近いことを明らかにした.

    脳波計測では、被験者心拍の収縮期と拡張期により、体性感覚誘発電位が変化することが知られているものの、心拍周期と外部刺激のタイミングによって生じる感覚野応答変化の詳細は未だよく分かっていない。そこで本研究は動物モデルを用いて脳神経活動を記録し、心拍動が神経活動を調節するメカニズムの解明を試みる。昨年度に引き続き、麻酔下ラットの心拍収縮期および拡張期に感覚刺激を与え、体性感覚野皮質に誘発される応答を局所フィールド電位計測法により解析する実験を行った。刺激トライアル毎の感覚応答のバラツキを解析した結果、収縮期における感覚応答のバラツキが収縮期のそれより大きいという現象が、実験個体数を増やしても再現されることが示された。一方で、膜電位イメージングを用いた感覚応答評価では、光応答振幅/空間的広がりの何れにおいても、心拍周期に依存する応答変化は検出できなかった。膜電位イメージング法は、心拍動に起因するアーティファクトを最小限に除去するようにプログラミングした信号処理を行うことから、本研究が対象とする心拍由来の応答変化検出には不向きである可能性が考えられた。また、本年度は、脳血管系への作用も指摘されている経頭蓋電気刺激(直流電気刺激)の組み合わせ効果についての実験も開始した。直流電気刺激が脳血管系制御を介して、心拍周期依存の感覚応答変化を調節することを検証する実験までは進めなかったものの、予備実験の段階で新しいタイプの神経可塑性現象を発見し論文発表を行った。

    The effects of pulsatile flow on brain parenchyma have not been well understood. Therefore, we examined the effect of pulsatile circulation on the brain using ex vivo whole brain preparations and an artificial blood circulation system. First, an experimental system was developed in which whole brain preparations could maintain physiological activity for 5 hours by pulsatile circulation of artificial blood. Next, electrical stimulation was given to the lateral olfactory tracts at different phases of the pulsatile flow to elicit neural activity, and the neural responses in the piriform cortex were analyzed. The results revealed a phenomenon in which local field potential activity changes relative to the phase of the pulsatile flow, suggesting that pulsatile circulation may modulate brain function.

    本研究では,嗜好性表出の脳内神経機構を,齧歯類の聴覚系に着目して明らかにすることを目指した.特に,聴覚入力に対して生じる「嗜好」が,知覚・記憶のプロセスを経て形成される「親密度」と結びつく可能性(後天的に獲得される嗜好)に着目し,神経行動・生理・解剖・薬理学的な手法を駆使して研究を進める計画とした.その第一歩として,ある種の音の「好み」が,生得的な要因により生じるものなのか,あるいは,後天的な要因によるものなのかという問題に取り組んだ.その際,動物の行動評価の際に用いる飲水報酬が,その後の選好行動に後天的な影響を与える可能性に着目して,行動実験パラダイムを構築した.ラットが,U字型ならびにM字型迷路下において,両端に提示される異なる音源を自発的に選択した行動を調査した結果,白色ノイズ音源とViolin音源には,ラットが生得的に好まない成分が含まれていることが示唆された.また,薬理学的解析,電気生理学的解析を行うための専用の行動実験系の構築に臨んだ.構築した実験系の評価を行うため,報酬待機行動課題を用いて,側坐核へのムシモルおよびドーパミン拮抗薬の注入実験を行い,それぞれの薬液が,ラットの待機行動に影響を及ぼすことが確認できた.また,「親密度」に関わる処理を担う可能性が示唆されている嗅周囲皮質について,聴覚野や前頭皮質と海馬との間に位置して,どのような役割を担っているのか?を脳スライスの膜電位イメージングにより解析し,in vivo行動下での神経活動の薬理阻害実験に活用するための知見を得た.

    To elucidate effects of dopaminergic and cholinergic activation on motor learning, we analyzed motor performance and cortical activity during electrical stimulation to the ventral tegmental area (VTA) or to the nucleus basalis of Meynert (NBM). Using the horizontal ladder task, it was found that VTA activation improved stepping performance (foot fault score) of rats. Transcranial direct current stimulation (tDCS) over the motor cortex restored the left-right motor imbalance in rat model of Parkinson’s disease. Furthermore, topographical mapping between the NBM and frontal cortex was determined, and electrical stimulation to the posterior part of the NBM was found to suppress excessive sensorimotor activity in the damaged brain. These findings may provide insight for future development of neurorehabilitation.

    A biomonitoring system aimed at assessment of lactic acid secretion from living organ sample was developed and utilized in brain lactic acid monitoring. Also, the biomonitoring system was tested in other applications. Using the biomonitoring system, (1) lactic acid produced in metabolic activity in block brain samples, (2) dependence of lactic acid secretion on the biochemical stimulation. (3) Also, our system was confirmed to be useful not only in the monitoring of organ level phenomenon, but also useful in monitoring of body fluids within individual-levels.

    The purpose of this study is to understand the emergent mechanism of "individuality" that guarantees the diversity of human behavior. In this project, we aim to develop measurement and evaluation techniques for brain circuit functions using experimental animals (mice and rats). The emergence of "individuality" depends on the function of large-scale, complex neural circuits consisting of many neurons and glial cells. To date, measurement methods of such neural circuit function is limited. In addition, the differences in the neural circuit functions responsible for the "individuality" are too small to detect and are easily hidden in the variability. Here, we have created a measurement technique using membrane voltage measurement with voltage-sensitive dyes to precisely capture the dynamics of large-scale neural circuit function.

    We aimed to identify the neural circuit, which elicited the preference behavior in rodents and to obtain findings which contribute to the understanding of its dynamics. Therefore, we designed the alternative selection task by using modified M-shaped-maze (70cm x 40cm x 45cm) to test the discrimination ability and the preference for different sound stimuli in rats and mice. Notably, we analyzed the behavior by focusing on the difference of acoustic stimulus in melody, tone, and tempo. As a result, 1) the rats could discriminate the type (timbre) of sound stimuli such as the difference among piano, violin, and bell, 2) specific animals exhibit preference behavior in which they chose the sound played by piano or violin more than the bell. Microinjection of muscimol (an agonist of GABAA receptor) during the behavioral experiment performed in the maze suggested that the orbitofrontal cortex and nucleus accumbens might contribute to the alternative selection behavior.

    This study aims to clarify how abnormality of E / I balance causes neuronal circuit information processing abnormality with voltage-sensitive dye (VSD) imaging. The precise activation control of E / I was involved in the short-term plasticity caused by the θ - γ cycle in the hippocampal CA 1 field. The observation was published as a paper in the first year. We have been proceeding with further verification in valproic acid administration model but no significant difference could be found with regard to this short term plasticity phenomenon with respect to this θ - γ cycle. Therefore, we focused on extraction of plastic change caused by this phenomenon and carried out research. I believe that basic data on the relationship of E / I balance disorder have been accumulated.

    How do the learning tasks inducing emotional responses cause the change of the brain function? For solving this problem, we focused on the fusional approach of animal behavioral analysis and ex vivo physiological analysis. We created model animals using a conditioned odor aversion (COA) learning paradigm. Additionally, exploratory surveys using voltage-sensitive dye (VSD) imaging of ex vivo brain preparations were performed. The visualization of the critical brain region for the learning behavior by applying the VSD imaging to brain preparations of COA model was unsuccessful. However, by applying VSD imaging to the olfactory cortices harvested from urethane-experienced animals, abnormal neural activity in response to electrical stimulation to the lateral olfactory tract was observed. This observation suggests that improvements in learning paradigm and recording procedures may enable us to measure the change of brain activities caused by behavioral changes controlled by the emotion.

    In this research, a high sensitive biosensing system suitable for multi-scale and multi-analyte biomonitoring. On the basis of biosensensors formed on the soft polymers, we utilized an Os-HRP modified carbon electrode, on which lactate oxidase was immobilized, formed on the PET sheet for biomonitoring purpose. The sensing system was utilized in the monitoring of lactic acid contained in the sweat and CSF, respectively. As a result, both lactic acid was successfully monitored using our system. The level of lactic acid in the sweat after anaerobic exercise increased by 7.2 folds in comparison with that of normal situation. This indicates that the biosensor reflects the physiological situation of the human subject.

    In order to reveal the functional organization of the amygdaloid network, we developed the optical recording system using a guinea pig isolated whole brain. By using this experimental setup, we analyzed the dependency of stimulus intensity and frequency on the spatiotemporal patterns of the neural activity evoked by the electrical shock to olfactory nerve. In addition to the imaging study, laminar profiles of evoked field potentials were recorded in the amygdala using a multichannel silicon probe. Current source density analysis performed on the field responses revealed the propagation pattern of the neural activity in the deep structure of the amygdaloid cortex.

    嗅覚情報伝達経路に関する知見を総合すると、嗅球-梨状皮質から、嗅内皮質を介して海馬へ入力する経路では「認識」に関する情報が、扁桃体周囲皮質を介して扁桃核に入力する経路では「情動」に関する情報が、それぞれ並列に処理されると考えられている。一方、これら二経路を結びつける回路については不明な点が多い。
    平成16年度の研究では、辺縁系神経回路の構造を保持したin vitro単離脳標本に膜電位イメージング手法を適用する事で、前述の二つの経路が外側嗅索の電気刺激により実際に活性化される事を確認した。又、実験の過程で、嗅内皮質で惹起された神経活動が海馬のみならず扁桃体周囲皮質へも伝達している可能性が示唆された。平成17年度は以下の実験により、扁桃体周囲皮質が嗅内皮質と機能的にリンクしている可能性を検証し、成果の一部を北米神経科学学会にて発表した。(1)局所的破壊が神経活動伝播パターンに及ぼす影響:梨状皮質外側部を破壊した場合、嗅内皮質への神経活動伝播が阻害されると共に、嗅内皮質から扁桃体周囲皮質への神経活動伝播も消失した。この時、梨状皮質-扁桃体周囲皮質間の神経活動伝播パターンに影響は無かった。又、梨状皮質内側部の破壊は、これと相反する結果を示した。(2)単一細胞記録解析:扁桃体周囲皮質の単一細胞内記録を行い、外側嗅索の電気刺激により惹起されるシナプス電位を解析した結果、多くの細胞のシナプス後電位は二峰性を示し、ピーク間隔は刺激強度に依存して変化する事が判った。細胞外記録電位及びイメージングの結果とも良く対応する事から、潜時の早いピークは嗅内皮質から、後発のピークは扁桃体周囲皮質からの神経支配に起因するものである事が推測された。
    これらの結果は、扁桃体周囲皮質が、梨状皮質からの嗅覚情報入力を受けると共に、嗅内皮質を介した高次の嗅覚情報も受け取り、統合している可能性を示唆している。
