High-intensity intermittent training (HIIT) is known to enhance mitochondrial biogenesis and energy metabolism in skeletal muscles. We aimed to elucidate the effects of HIIT on mitochondrial dynamics proteins, which is coupled with mitochondrial morphological adaptation. ICR mice (male, 8 weeks old) were divided into sedentary or HIIT groups (n=6 per group). The HIIT group underwent 10-12 sets of swimming for 20 s per set and inter-set intervals of 10 s, with a load of 10% body weight. Training sessions were conducted five times per week, and each training period lasted for 6 weeks. Gastrocnemius muscles were used for western blotting analysis. HIIT led to an increase in the expression of PGC-1αand OXPHOS proteins; however, Com- plex2 expression did not change. HIIT group showed higher ex- pressions of the mitochondrial fusion proteins Mfn2 and Opa1 and the mitochondrial fission protein Drp1 compared with that of the Sed group. The findings of this study indicate that HIIT enhances both mitochondrial biogenesis and mitochondrial dynamics-relat- ed proteins.

<p>Eicosapentaenoic acid (EPA) is an omega-3 polyunsaturated fatty acid that is abundant in fish oil, and has anti-inflammatory or anti-obesity effects. However, the effects of EPA supplementation on mitochondrial content and dynamics (fusion and fission) in skeletal muscle has not been elucidated. We investigated the effects of EPA intake for 4 weeks on denervation-induced mitochondrial adaptation in mice skeletal muscle. ICR mice (male, 8 weeks old) were daily administrated olive oil (control) or EPA at a dose of 300 mg/kg body weight by gavage for 4 weeks. After 2 weeks of oil intake, mice underwent unilateral sciatic nerve transection surgery. The hindlimb without surgery served as the sham-operated control. Body and skeletal muscle weights did not differ between the control and the EPA groups. Peroxisome proliferator-activated receptor gamma coactivator 1 alpha (PGC-1α) and oxidative phosphorylation (OXPHOS) proteins were significantly decreased by denervation surgery. Denervation also induced the reduction of mitochondrial fusion molecules and the increase of mitochondrial fission molecules. Expression levels of PGC-1α and mitochondrial respiration protein MTCO1 were higher

Delay in the growth of skeletal muscle resulting from physical inactivity during the juve- nile period may affect the subsequent quality of life. Therefore, it is important to clarify whether recovery of this muscle growth de- lay can be achieved by physical training thereafter. In the present study, mice were subjected to hindlimb suspension from 5 to 8 weeks of age to induce a delay of muscle growth and reloading from 8 weeks of age to recovery of muscle. Growth of the planta- ris muscle, which is composed of fast-twitch fibers, was monitored at 5, 8, 11 and 14 weeks of age. This revealed that the growth of the plantaris had completely recovered from this delay by 11 weeks of age. Moreover, the recovery was accompanied by a sig- nificant increase of RNA content relative to control muscle. Since ribosomal RNA accounts for the majority of total RNA, it was thought that the machinery for protein synthesis had become acti- vated in the recovering muscle. In addition, phosphorylated ribo- somal protein S6, which is an important factor for efficient trans- lation, was increased at the same time. These results suggested that protein synthesis was enhanced in recovering muscle. In addi-
